What is NPXL?
NPXL is a medication used to treat erectile dysfunction. It is a phosphodiesterase type 5 inhibitor (PDE5 inhibitor). NPXL is used to improve blood flow to the penis during sexual stimulation. This helps men to get and maintain an erection. NPXL does not protect against sexually transmitted diseases (STDs).
How to take NPXL
Follow all directions on your prescription label. Do not take this medicine in larger or smaller amounts or for longer than recommended. NPXL is usually taken only when needed, about 30 minutes to 1 hour before sexual activity. You may take NPXL with or without food. NPXL can be taken with or without food. If you take NPXL with certain other medicines, the effects of either could be increased, decreased, or altered. An interaction between two medications does not always mean that you must stop taking one of the medications; however, sometimes it does. Tell your doctor about all other medications you use for erectile dysfunction.
Side effects of NPXL
Get emergency medical help if you have signs of an allergic reaction: hives; difficulty breathing; swelling of your face, lips, tongue, or throat. Call your doctor at once if you have:
- a light-headed feeling, like you might pass out;
- an erection that is painful or lasts 4 hours or longer;
- a sudden decrease or loss of hearing; or
- a sudden decrease or loss of vision.
NPXL can decrease blood flow to the optic nerve of the eye, causing sudden vision loss. This has occurred in a small number of people taking PDE5 inhibitors, including NPXL. It is not clear whether NPXL is the actual cause of vision loss.
Stop taking NPXL and get emergency medical help if you have sudden vision loss.
During sexual activity, if you become dizzy or nauseated, or have pain, numbness, or tingling in your chest, arms, neck, or jaw, stop and call your doctor right away.
You could be having a serious side effect of NPXL.
Stop taking NPXL and call your doctor at once if you have:
- a feeling like you might pass out;
- ringing in your ears, or sudden hearing loss;
- chest pain or heavy feeling, pain spreading to the arm or shoulder, nausea, sweating, general ill feeling;
- irregular heartbeat;
- swelling in your hands, ankles, or feet;
- shortness of breath;
- vision changes or sudden vision loss; or
- feeling light-headed, fainting.
Common NPXL side effects may include:
- flushing (warmth, redness, or tingly feeling);
- headache, dizziness;
- abnormal vision (blurred vision, changes in color vision) ;
- runny or stuffy nose, nosebleeds;
- sleep problems (insomnia);
- muscle pain, back pain; or
- upset stomach.

NPXL dosing information
Usual Adult Dose for Erectile Dysfunction:
- Initial dose: 50 mg orally once a day, as needed, 1 hour prior to sexual activity
- Maintenance: 25 to 100 mg orally once a day, as needed, 1 hour prior to sexual activity
- Maximum dose: 100 mg orally once a day
Comments:
- Sexual stimulation is required for a response to treatment.
- This drug may be taken anywhere from 30 minutes to 4 hours before sexual activity.
- NPXL should not be taken more than once a day.

- Treatment of erectile dysfunction
Limitations of use:
- NPXL is not for use in women
What other drugs will affect NPXL?
- Tell your doctor about all other medications you use for erectile dysfunction.
Using NPXL with other drugs may cause serious side effects. Do not take NPXL with similar medications such as avanafil (Stendra), sildenafil (Viagra), or tadalafil (Cialis).
Ask your doctor before taking a sleeping pill, narcotic medication, muscle relaxer, or medicine for anxiety, depression, or seizures.
Tell your doctor about all your current medicines and any you start or stop using, especially:
- drugs to treat high blood pressure or a prostate disorder;
- an antibiotic - clarithromycin, erythromycin, or rifampin;
- antifungal medicine - ketoconazole or itraconazole; or
- HIV/AIDS medication - atazanavir, indinavir, ritonavir, or saquinavir.
